    A histopathological evaluation was conducted on the kidneys of mice following their sacrifice two days after an intravenous injection with 1 × 106 C. albicans SC5314 cells, administered with or without 2 mg/kg CaHsp90 (n = 3). The kidneys were harvested, and two consecutive sections were stained using PAS stain and IHC staining with an anti-IL-1β antibody, respectively.

    J774A.1 cells were pre-incubated in the presence or absence of CaHsp90 at concentrations of 0.0625 μM, 0.25 μM, and 1 μM for 4 hours. Following this pre-incubation, the cells were exposed to C. albicans SC5314 cells (MOI = 10) for an additional 12 hours. Subsequently, the protein levels of IL-1β in the supernatant were analyzed using Western blotting.

    • Western blot analysis of extracts from A431(lane 2(20μg), RAW264.7 (lane 3(20μg) and A549(lane 4(20μg) using IL-1 beta (HY-P80720) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

    • Immunocytochemistry analysis of RAW 264.7 cells labeling IL-1 beta with IL-1 beta Antibody (HY-P80720)at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with IL-1 beta Antibody (HY-P80720) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L(HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

    Function:Potent pro-inflammatory cytokine (PubMed:10653850, PubMed:12794819, PubMed:28331908, PubMed:3920526). Initially discovered as the major endogenous pyrogen, induces prostaglandin synthesis, neutrophil influx and activation, T-cell activation and cytokine production, B-cell activation and antibody production, and fibroblast proliferation and collagen production (PubMed:3920526). Promotes Th17 differentiation of T-cells. Synergizes with IL12/interleukin-12 to induce IFNG synthesis from T-helper 1 (Th1) cells (PubMed:10653850). Plays a role in angiogenesis by inducing VEGF production synergistically with TNF and IL6 (PubMed:12794819). Involved in transduction of inflammation downstream of pyroptosis: its mature form is specifically released in the extracellular milieu by passing through the gasdermin-D (GSDMD) pore (PubMed:33377178, PubMed:33883744). Acts as a sensor of S.pyogenes infection in skin: cleaved and activated by pyogenes SpeB protease, leading to an inflammatory response that prevents bacterial growth during invasive skin infection (PubMed:28331908)
