Western blot analysis of extracts from HeLa (lane 1(20μg)) 、HepG2 (lane 2(20μg)) 、Mouse heart (lane 3(20μg)) and Rat heart (lane 4(20μg)) using HADH Antibody (HY-P86895) . Proteins were transferred to a PVDF membrane and blocked with 5% nonfat dry milk in TBST for 1.5 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (β Tubulin, 1/10000) was used in 5% nonfat dry milk in TBST at 4℃ overnight. Goat Anti-Mouse IgG-HRP Secondary Antibody (1/10,000) was used for 1 hour at room temperature.
Background

Function
HADH is a Mitochondrial fatty acid beta-oxidation enzyme that catalyzes the third step of the beta-oxidation cycle for medium and short-chain 3-hydroxy fatty acyl-CoAs (C4 to C10). Plays a role in the control of insulin secretion by inhibiting the activation of glutamate dehydrogenase 1 (GLUD1), an enzyme that has an important role in regulating amino acid-induced insulin secretion. Plays a role in the maintenance of normal spermatogenesis through the reduction of fatty acid accumulation in the testes[1][2][3].
