  • Experimental Validation Results for Cofilin Antibody (YA2822)
Western blot analysis was performed on extracts from Hela (lane 1, 20 μg), and C6 (lane 2, 20 μg) using Cofilin Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non - fat milk in TBST at 4°C overnight. The primary antibody (1:1000 dilution) and the loading control antibody (beta-Tubulin, HY-P80955, 1:5000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti - Rabbit/Mouse IgG - HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.

Background

  • Function

Cofilin binds to F-actin and exhibits pH-sensitive F-actin depolymerizing activity. In conjunction with the subcortical maternal complex (SCMC), plays an essential role for zygotes to progress beyond the first embryonic cell divisions via regulation of actin dynamics. Required for the centralization of the mitotic spindle and symmetric division of zygotes. Plays a role in the regulation of cell morphology and cytoskeletal organization in epithelial cells. Required for the up-regulation of atypical chemokine receptor ACKR2 from endosomal compartment to cell membrane, increasing its efficiency in chemokine uptake and degradation. Required for neural tube morphogenesis and neural crest cell migration[1][2][3][4].

  • Subcellular Localization

Nucleus matrix; Cytoplasm, cytoskeleton; Cell projection, ruffle membrane; Peripheral membrane protein; Cytoplasmic side; Cell projection, lamellipodium membrane; Peripheral membrane protein; Cytoplasmic side; Cell projection, lamellipodium; Cell projection, growth cone; Cell projection, axon

  • Expression

    Tissue_specificity:Widely distributed in various tissues

Induction:Up-regulated in response to enterovirus 71 (EV71) infection (at protein level)

  • Subunit

Can bind G- and F-actin in a 1:1 ratio of cofilin to actin (PubMed:11812157). It is a major component of intranuclear and cytoplasmic actin rods (By similarity). Interacts with the subcortical maternal complex (SCMC) via interaction with TLE6 isoform 1 and NLRP5 (By similarity). Interacts with C9orf72 (By similarity)
