Background

Function
XRCC1 is a Scaffold protein involved in DNA single-strand break repair by mediating the assembly of DNA break repair protein complexes. Negatively regulates ADP-ribosyltransferase activity of PARP1 during base-excision repair in order to prevent excessive PARP1 activity. Recognizes and binds poly-ADP-ribose chains: specifically binds auto-poly-ADP-ribosylated PARP1, limiting its activity[1][2][3][4][5].

-
Expression
Tissue_specificity:It is expressed (at the protein level) in fibroblasts, retinal pigment epithelial cells, and lymphoblasts. -
Subunit
Homodimer (PubMed:16397295). Interacts with polynucleotide kinase (PNK), DNA polymerase-beta (POLB) and DNA ligase III (LIG3) (PubMed:19155274, PubMed:19336415). Interacts with APTX and APLF (PubMed:14755728, PubMed:15044383, PubMed:15380105, PubMed:17353262, PubMed:17507382). Interacts with APEX1; the interaction is induced by SIRT1 and increases with the acetylated form of APEX1 (PubMed:19934257). Interacts with (poly-ADP-ribosylated) PARP1 (PubMed:34811483)
