Western blot analysis of extracts from A431(lane 2(20ug) and A431(lane 3(40ug) using Integrin alpha 6 Antibody (HY-P83275) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P83730, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

Integrin alpha 6 is an Integrin alpha-6/beta-1 (ITGA6:ITGB1) is a receptor for laminin on platelets. Integrin alpha-6/beta-1 (ITGA6:ITGB1) is present in oocytes and is involved in sperm-egg fusion. Integrin alpha-6/beta-4 (ITGA6:ITGB4) is a receptor for laminin in epithelial cells and it plays a critical structural role in the hemidesmosome. ITGA6:ITGB4 binds to NRG1 (via EGF domain) and this binding is essential for NRG1-ERBB signaling. ITGA6:ITGB4 binds to IGF1 and this binding is essential for IGF1 signaling. ITGA6:ITGB4 binds to IGF2 and this binding is essential for IGF2 signaling[1][2][3].

Tissue_specificity:Integrin α6/β4 is primarily expressed by epithelial cells. The isoform containing the X1 fragment is universally expressed. The isoform containing the X1X2 fragment is expressed in the heart, kidney, placenta, colon, duodenum, myoblasts and myotubes, and a few cell lines; they are always co-expressed with the universally present isoform containing the X1 fragment. In some tissues (e.g., salivary glands) , isoforms containing both cytoplasmic fragment A and fragment B can be detected, while in others, only an isoform containing one cytoplasmic fragment is found (fragment A in the epidermis, fragment B in the kidneys) . Processed integrin α6: expressed at low levels in normal prostate tissue, with elevated levels in prostate cancer tissue (protein level) (PubMed: 15023541) . -
