Western blot analysis of extracts from Jurkat (lane2, 20μg) using Wnt2 Antibody (HY-P83228). Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80993, 1/10,000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (HY-P8001,1/10,000) was used for 1 hour at room temperature.

Function
Wnt2 functions within the Wnt/β-catenin signaling system, a conserved pathway that regulates development, adult tissue stem cells, and growth-related disease processes[1]. Mechanistically, Wnt2/2b signaling through canonical β-catenin specifies lung endoderm progenitors in the anterior foregut, and pathway activation can reprogram esophagus and stomach endoderm toward lung progenitor fate[2]. In lung mesenchyme, Wnt2 activates the airway smooth muscle program by regulating myocardin/Mrtf-B and Fgf10 expression[3]. In colorectal cancer cells, Wnt2 is highly expressed, co-expressed with Wnt/β-catenin target genes, and required for target-gene expression and cell proliferation[4]. Compared with related isoforms, WNT2B is a WNT2 paralogue that encodes WNT2B1 and WNT2B2 through alternative splicing and alternative promoter usage[5]. For experimental applications, Wnt2 loss-of-function, Wnt2 neutralization, β-catenin deletion, Dkk1 expression, and multi-Frizzled antibody blockade provide practical approaches to test ligand-dependent Wnt signaling in developmental and cancer models[2][4][6].
