Background

  • Function

Collagen II is the major collagen synthesized by chondrocytes and a core structural component of articular cartilage, vitreous humour, inner ear, and nucleus pulposus[1]. Mechanistically, COL2A1 suppresses articular chondrocyte hypertrophy and osteoarthritis progression by promoting integrin β1-SMAD1 interaction and regulating BMP-SMAD1 signaling[2]. In disease models, COL2A1 loss accelerates chondrocyte hypertrophy, while degenerative human articular cartilage shows reduced COL2A1 with increased hypertrophic markers and BMP-SMAD1 activity[2]. Type II collagen degradation can also expose a cryptic peptide sequence that induces collagenase activity, MMP-13, and chondrocyte hypertrophy in articular cartilage explants[3]. Compared with type X collagen, which marks hypertrophic chondrocytes in osteoarthritis, collagen II better reflects the stable hyaline cartilage phenotype[4]. Compared with collagen I devices, collagen II biomaterials more directly match cartilage extracellular matrix composition and support cartilage engineering research[5]. For experimental applications, collagen II scaffolds help maintain or rejuvenate hyaline chondrocyte phenotype through Wnt-related mechanisms, supporting use in cartilage tissue engineering models[6].

Tissue_specificity:Isomer 2 is highly expressed in juvenile chondrocytes and lowly expressed in fetal chondrocytes.

  • Isoforms & Post-Translational Modification

    P02458 has 3 isomers: P02458-2: 141785 Da (predicted); P02458-1: 134389 Da (predicted); P02458-3: 29781 Da (predicted).
The N-telopeptide is covalently linked to the helical COL2 region of alpha 1(IX), alpha 2(IX) and alpha 3(IX) chain. The C-telopeptide is covalently linked to an another site in the helical region of alpha 3(IX) COL2;Contains mostly 4-hydroxyproline. Prolines at the third position of the tripeptide repeating unit (G-X-P) are 4-hydroxylated in some or all of the chains;Contains 3-hydroxyproline at a few sites. This modification occurs on the first proline residue in the sequence motif Gly-Pro-Hyp, where Hyp is 4-hydroxyproline;Lysine residues at the third position of the tripeptide repeating unit (G-X-Y) are 5-hydroxylated in some or all of the chains;O-glycosylated on hydroxylated lysine residues. The O-linked glycan consists of a Glc-Gal disaccharide
