Western blot analysis was performed on extracts from Hela (lane 1, 15 μg), and U2OS (lane 2, 15 μg) using Integrin Linked ILK Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Actin, HY-P83730, 1:20000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
Western blot analysis of extracts from Hela (lane 2(20μg), Hela (lane 3(40μg), using Integrin Linked ILK Antibody. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.

Background

Function：Scaffold protein which mediates protein-protein interactions during a range of cellular events including focal adhesion assembly, cell adhesion and cell migration (PubMed:17420447, PubMed:20005845, PubMed:30367047, PubMed:32528174). Regulates integrin-mediated signal transduction by contributing to inside-out integrin activation (By similarity). Recruits PARVA and LIMS1/PITCH to form the heterotrimeric IPP (ILK-PINCH-PARVIN) complex which binds to F-actin via the C-terminal tail of LIMS1 and the N-terminal region of PARVA, promoting F-actin filament bundling, a process required to generate force for actin cytoskeleton reorganization and subsequent dynamic cell adhesion events such as cell spreading and migration (PubMed:30367047). Binding to PARVA promotes effective assembly of ILK into focal adhesions while PARVA-bound ILK can simultaneously engage integrin-beta cytoplasmic tails to mediate cell adhesion (PubMed:20005845). Plays a role with PARVG in promoting the cell adhesion and spreading of leukocytes (PubMed:16517730). Acts as an upstream effector of both AKT1/PKB and GSK3 (PubMed:9736715). Mediates trafficking of caveolae to the cell surface in an ITGB1-dependent manner by promoting the recruitment of IQGAP1 to the cell cortex which cooperates with its effector DIAPH1 to locally stabilize microtubules and allow stable insertion of caveolae into the plasma membrane (By similarity). Required for the maintenance of mitotic spindle integrity by promoting phosphorylation of TACC3 by AURKA (PubMed:18283114). Associates with chromatin and may act as a negative regulator of transcription when located in the nucleus (PubMed:17420447)

Subunit：Component of the heterotrimeric IPP (ILK-PINCH-PARVIN) complex composed of ILK, LIMS1/PINCH and PARVA; the complex binds to F-actin via the C-terminal tail of LIMS1 and the N-terminal region of PARVA, promoting F-actin filament bundling (PubMed:11331308, PubMed:12167643, PubMed:12432066, PubMed:19074270, PubMed:19117955, PubMed:21524996, PubMed:30367047, PubMed:35259013). Formation of the IPP complex is dependent on protein kinase C and precedes integrin-mediated cell adhesion and spreading (PubMed:12432066). ILK also interacts with LIMS2/PINCH2 and with PARVB and PARVG which may substitute for LIMS1 and PARVA in the IPP complex; PARVA and PARVB compete for the same binding site (PubMed:11402068, PubMed:12167643, PubMed:15284246, PubMed:15817463, PubMed:16517730, PubMed:20005845, PubMed:20117114). Interaction with PARVG promotes the establishment of cell polarity required for leukocyte migration (PubMed:16517730). Interacts with the cytoplasmic domain of integrin ITGB1 and may also interact with integrins ITGB2, ITGB3 and/or ITGB5 (PubMed:8538749). Interacts probably also with TGFB1I1 (By similarity). Interacts (via ANK repeats) with EPHA1 (via SAM domain); stimulated by EFNA1 but independent of the kinase activity of EPHA1 (PubMed:19118217). Interacts with FERMT2 (By similarity). Interacts with LIMD2; leading to activate the protein kinase activity (PubMed:24590809). Interacts with PXN/PAXILLIN (via LD motif 4) (PubMed:15817463).
