Western blot analysis was performed on extracts from CHO-K1 (lane 1, 15 μg), CHO-K1(EGFP-EIF4EBP3) (lane 2, 15 μg) using 4E BP3 (3B6) Mouse mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non - fat milk in TBST at 4°C overnight. The primary antibody (1:1000 dilution) and the loading control antibody (beta-Actin(HRP), HY-P80993, 1:5000 dilution) was incubated in 5% non-fat milk in TBST for 1 hour at 37°C. Goat Anti - Mouse IgG - HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.

eIF4EBP3 is a Repressor of translation initiation that regulates EIF4E activity by preventing its assembly into the eIF4F complex: the hypophosphorylated form competes with EIF4G1/EIF4G3 and strongly binds to EIF4E, leading to repression of translation. In contrast, the hyperphosphorylated form dissociates from EIF4E, allowing interaction between EIF4G1/EIF4G3 and EIF4E, leading to initiation of translation. Inhibits EIF4E-mediated mRNA nuclear export[1].
