Western blot analysis was performed on protein extracts (25 μg) from Rat skin (lane 2, Negative), Mouse skin (lane 3), Rat stomach (lane 4), HeLa (lane 5), A549 (lane 6), and THP-1 (lane 7) using Cytokeratin 10 antibody. Proteins were transferred onto a 0.45 μm PVDF membrane using the Trans-Blot® Turbo™ system for 13 min. The membrane was then blocked with 5% nonfat milk in TBST (HY-K1025) for 1 h at room temperature. The primary antibody (1:1000) and loading control antibody GAPDH Antibody (HRP) (HY-P80954A) (1:2500) were diluted in 5% nonfat milk in TBST and incubated with the membrane overnight at 4°C. After washing, the membrane of primary antibody was incubated with HRP-conjugated goat anti-rabbit/mouse IgG secondary antibody (HY-P8001/HY-P8004) (1:5000) diluted in 5% nonfat milk in TBST for 1 h at room temperature. Protein bands were visualized using an Ultra High Sensitivity ECL detection kit (HY-K1005).

Cytokeratin 10 plays a role in the establishment of the epidermal barrier on plantar skin. Involved in the maintenance of cell layer development and keratin filament bundles in suprabasal cells of the epithelium; (Microbial infection) Acts as a mediator of S.aureus adherence to desquamated nasal epithelial cells via clfB, and hence may play a role in nasal colonization; (Microbial infection) Binds S.pneumoniae PsrP, mediating adherence of the bacteria to lung cell lines. Reduction of levels of KRT10 keratin decrease adherence, overexpression increases adherence. Neither protein has to be glycosylated for the interaction to occur

Tissue_specificity:It can be found in all cell layers above the basal layer, including the stratum corneum. It is expressed on the surface of lung cell lines (PubMed:19627498) . It is localized on the surface of exfoliated nasal epithelial cells (protein level) (PubMed:12427098) .
Induction:Repressed in keratinocytes by all-trans retinoic acid (ATRA) , via reduction of mRNA stability -

Heterotetramer of two type I and two type II keratins. Heterodimer with KRT1 (PubMed:24940650, PubMed:27595935). Two heterodimers of KRT1 and KRT10 form a heterotetramer (PubMed:27595935). The KRT10 subunit in the heterotetramer is probably disulfide-linked (PubMed:27595935). Interacts with PLEC isoform 1C, when in a heterodimer with KRT1 (PubMed:24940650)
