Western blot analysis was performed on extracts from NCCIT (lane 1, 15 μg) using Cripto 1 Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight. The primary antibody (1:1000 dilution) and the loading control antibody (GAPDH, HY-P80137, 1:80000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
Background

Function
GPI-anchored cell membrane protein involved in Nodal signaling. Cell-associated CRIPTO acts as a Nodal coreceptor in cis. Shedding of CRIPTO by TMEM8A modulates Nodal signaling by allowing soluble CRIPTO to act as a Nodal coreceptor on other cells (PubMed:27881714). Could play a role in the determination of the epiblastic cells that subsequently give rise to the mesoderm (PubMed:11909953)

Subcellular Localization
Cell membrane; Lipid-anchor, GPI-anchor; Secreted
-
Expression
Tissue_specificity:This gene is preferentially expressed in gastric and colorectal cancers compared to normal tissues. It is also expressed in breast and lung cancers. -
Subunit
Interacts with the activin type-1 receptor ACVR1B
