WB
  • Western blot analysis was performed on extracts from Jurkat (lane 1, 20 μg), Hela (lane 2, 20 μg), Ramos (lane 3, 20 μg), and A431 (lane 4, 20 μg) using Cyclin E2 Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non - fat milk in TBST at 4°C overnight. The primary antibody (1:1000 dilution) and the loading control antibody (beta-Actin, HY-P83730, 1:5000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti - Rabbit IgG - HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.

Background
Function:Essential for the control of the cell cycle at the late G1 and early S phase
Subcellular Localization:Nucleus
Expression:
Tissue_specificity:According to PubMed:9858585, highest levels of expression in adult testis, thymus and brain. Lower levels in placenta, spleen and colon. Consistently elevated levels in tumor-derived cells compared to non-transformed proliferating cells. According to PubMed:9840927: low levels in thymus, prostate, brain, skeletal muscle, and kidney. Elevated levels in lung. According to PubMed:9840943 highly expressed in testis, placenta, thymus and brain. In a lesser extent in small intestine and colon

Induction:Activated by papilloma viral oncoproteins E6 and E7 which bind to and inactivate p53 and Rb, respectively
Isoforms & Post-Translational Modification:O96020 has 2 isomers: O96020-1: 46757 Da (predicted); O96020-2: 41465 Da (predicted).
Phosphorylation by CDK2 triggers its release from CDK2 and degradation via the ubiquitin proteasome pathway;Lactylated at Lys-348 (PubMed:36896611). Delactylated by SIRT3 (PubMed:36896611)
Subunit:Interacts with the CDK2 (in vivo) and CDK3 (in vitro) protein kinases to form a serine/threonine kinase holoenzyme complex. The cyclin subunit imparts substrate specificity to the complex

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
