    Low-density lipoprotein receptor-related protein 2 (LRP2), also known as megalin, is an endocytic receptor expressed on the apical surface of epithelial cells that internalizes diverse ligands, including nutrients, hormones, morphogens, and extracellular matrix proteins[1]. Mechanistically, LRP2 modulates sonic hedgehog (SHH) signaling in stem and progenitor cell niches, playing crucial roles in brain, eye, and heart development[2]. This receptor participates in both lysosomal degradation and transcytosis, with its surface availability regulated by phosphorylation, extracellular domain shedding, intramembrane proteolysis, and exosome secretion[1]. LRP2 dysfunction has been linked to congenital syndromes, diabetic nephropathy, Dent disease, Lowe syndrome, Alzheimer's disease, and neural regeneration deficits[1][2]. Compared with related LDL receptor family members, LRP2 uniquely acts as an auxiliary receptor for SHH, whereas LRP5/6 primarily function as Wnt co-receptors and LRP1 modulates inflammatory signaling[3][2][4].
