Western blot analysis of extracts from HT-29 (lane2(20μg), A549 (lane3(20μg), HepG2 (lane4(20μg) and PC-3 (lane5(20μg) using FoxA2 Antibody (HY-P84884). Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80993, 1/10,000) was used in 5% non-fat milk in TBST for 2 hour at room temperature. Goat Anti-Mouse IgG-HRP Secondary Antibody (HY-P8004, 1/10,000) was used for 1 hour at room temperature.

Function
FOXA2 belongs to FOXA protein family. During pancreatic genealogy specification, FOXA2 is required to establish the initiation enhancer state and the correct remodeling of chromatin structure to recruit additional transcription factors, such as GATA6, for the activation of the transcriptional program required for human pancreatic differentiation. FOXA2 particularly affects the transition from the hind foregut (FG) to the primary pancreatic progenitor (PP1) stage. Mice with specific deletion of endodermal FOXA2 also exhibited normal growth of the liver. Consistent with the reduction in glucagon gene expression levels, the content of glucagon was reduced in FOXA1-, FOXA2-, and FOXA1/FOXA2-deficient α cells, and it seems that only the combined silencing of FOXA1 and FOXA2 can affect the secretion of glucagon[1].
