    Western blot analysis of extracts from SMMC-7721(lane 1(40μg) ),Jukat(lane 2(40μg) ),293T(lane 3(40μg) ),MCF-7(lane 4(40μg) )and Hela(lane 5(40μg) ) using KLF15 antibody. Proteins were transferred to a NC membrane and blocked with 5% Skim milk in TBST for 2 hour at room temperature. The primary antibody ( 1/1000) and Loading control antibody (GAPDH,1/1000) was used in 5% Skim milk in TBST at 4°C overnight. Goat Anti-Mouse IgG-HRP Secondary Antibody (1/10,000) was used for 1 hour at room temperature.
  • Experimental Validation Results for KLF15 Antibody (YA4561)
    Western blot analysis was performed on protein extracts (25 μg) from HeLa (lane 2), Ramos (lane 3), HepG2 (lane 4), SH-SY5Y (lane 5), and K562 (lane 6) using KLF15 antibody. Proteins were transferred onto a 0.45 μm PVDF membrane using the Trans-Blot® Turbo system for 13 min. The membrane was then blocked with 5% nonfat milk in TBST (HY-K1025) for 1 h at room temperature. The primary antibody (1:1000) and loading control antibody GAPDH Antibody (HRP) (HY-P80954A) (1:5000) were diluted in 5% nonfat milk in TBST and incubated with the membrane overnight at 4°C. After washing, the membrane of primary antibody was incubated with HRP-conjugated goat anti-rabbit/mouse IgG secondary antibody (HY-P8001/HY-P8004) (1:5000) diluted in 5% nonfat milk in TBST for 1 h at room temperature. Protein bands were visualized using an Ultra High Sensitivity ECL detection kit (HY-K1005).

    Flow cytometric analysis of 1X10^6 Ramos cells labeling KLF15 Antibody(red). Cells were fixed with 4% paraformaldehyde and permeabilised with 0.2% Triton X-100. Then stained with the primary antibody at 1/400 dilution overnight at 4℃. AF 488 Goat Anti-mouse IgG H&L was used as the secondary antibody at 1/1,000 dilution for 45 minutes at room temperature. Mouse IgG Isotype Control (blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).

    KLF15 transcriptional regulator that binds to the GA element of the CLCNKA promoter. Binds to the KCNIP2 promoter and regulates KCNIP2 circadian expression in the heart. Is a repressor of CCN2 expression, involved in the control of cardiac fibrosis. It is also involved in the control of cardiac hypertrophy acting through the inhibition of MEF2A and GATA4. Involved in podocyte differentiation. Inhibits MYOCD activity. Is a negative regulator of TP53 acetylation. Inhibits NF-kappa-B activation through repression of EP300-dependent RELA acetylation

    Tissue_specificity:It is highly expressed in the liver, skeletal muscle, and kidneys. It is also expressed in cardiomyocytes. Expression is significantly reduced in cardiac tissues with non-ischemic cardiomyopathy and aortic aneurysms, as well as in glomerular diseases. It is not expressed in bone marrow or lymphoid tissues.
