Western blot analysis was performed by loading 0.5 μg (Lane 2), 1 μg (Lane 3), and 2 μg (Lane 4) of MLANA Protein, Human (His, B2M) using Melan-A antibody. Proteins were transferred onto a 0.45 μm PVDF membrane using the Trans-Blot® Turbo™ system for 13 min. The membrane was then blocked with 5% nonfat milk in TBST (HY-K1025) for 1 h at room temperature. The primary antibody (1:1000) was diluted in 5% nonfat milk in TBST and incubated with the membrane overnight at 4°C. After washing, the membrane of primary antibody was incubated with HRP-conjugated goat anti-rabbit/mouse IgG secondary antibody (HY-P8001/HY-P8004) (1:5000) diluted in 5% nonfat milk in TBST for 1 h at room temperature. Protein bands were visualized using an Ultra High Sensitivity ECL detection kit (HY-K1005).
Background
-
Function
Melan-A is involved in melanosome biogenesis by ensuring the stability of GPR143. Plays a vital role in the expression, stability, trafficking, and processing of melanocyte protein PMEL, which is critical to the formation of stage II melanosomes
-
Subcellular Localization
Endoplasmic reticulum membrane; Single-pass type III membrane protein; Golgi apparatus; Golgi apparatus, trans-Golgi network membrane; Melanosome
-
Expression
Tissue_specificity:This gene is expressed only in melanoma and melanocyte lines, as well as in the retina. -
Subunit
Interacts with PMEL. Interacts with GPR143
