Background
Function:Protein tyrosine-protein phosphatase required for T-cell activation through the antigen receptor (PubMed:35767951). Acts as a positive regulator of T-cell coactivation upon binding to DPP4. The first PTPase domain has enzymatic activity, while the second one seems to affect the substrate specificity of the first one. Upon T-cell activation, recruits and dephosphorylates SKAP1 and FYN. Dephosphorylates LYN, and thereby modulates LYN activity (By similarity). Interacts with CLEC10A at antigen presenting cell-T cell contact; CLEC10A on immature dendritic cells recognizes Tn antigen-carrying PTPRC/CD45 receptor on effector T cells and modulates T cell activation threshold to limit autoreactivity; (Microbial infection) Acts as a receptor for human cytomegalovirus protein UL11 and mediates binding of UL11 to T-cells, leading to reduced induction of tyrosine phosphorylation of multiple signaling proteins upon T-cell receptor stimulation and impaired T-cell proliferation
Subcellular Localization:Cell membrane; Single-pass type I membrane protein; Membrane raft; Synapse
Expression:
Tissue_specificity:Isomer 1: Detected in thymocytes. Isomer 2: Detected in thymocytes. Isomer 3: Detected in thymocytes. Isomer 4: Not detected in thymocytes. Isomer 5: Detected in thymocytes. Isomer 6: Not detected in thymocytes. Isomer 7: Detected in thymocytes. Isomer 8: Not detected in thymocytes.
Isoforms & Post-Translational Modification:P08575 has 8 isomers: P08575-3: 147486 Da (predicted); P08575-4: 131130 Da (predicted); P08575-5: 142623 Da (predicted); P08575-6: 142898 Da (predicted); P08575-7: 140582 Da (predicted); P08575-8: 138034 Da (predicted); P08575-9: 135719 Da (predicted); P08575-10: 135993 Da (predicted).
Heavily N- and O-glycosylated
Subunit:Binds GANAB and PRKCSH (By similarity). Interacts with SKAP1 (PubMed:11909961). Interacts with DPP4; the interaction is enhanced in an interleukin-12-dependent manner in activated lymphocytes (PubMed:12676959)

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
