Western blot analysis was performed on extracts from U2OS (lane 1, 20 μg), 3T3 (lane 2, 20 μg), and Raji (lane 3, 20 μg) using PDL2 Mouse mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non - fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Actin,HY-P83730, 1:5000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti - Mouse/Rabbit IgG - HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.

Background

Function：Involved in the costimulatory signal, essential for T-cell proliferation and IFNG production in a PDCD1-independent manner. Interaction with PDCD1 inhibits T-cell proliferation by blocking cell cycle progression and cytokine production (By similarity)

Expression：
Tissue_specificity:It is highly expressed in the heart, placenta, pancreas, lungs, and liver, and lowly expressed in the spleen, lymph nodes, and thymus.

Induction:Up-regulated by IFNG/IFN-gamma stimulation in monocytes and induced on dendritic cells grown from peripheral blood mononuclear cells with CSF2 and IL4/interleukin-4
