Western blot analysis of extracts from THP-1 (lane 2(20μg) and THP-1 (lane 3(40μg)using CD31 (HY-P80599) Mouse mAb. Proteins were transferred to a PVDF membrane and blocked with 5% defatted milk powder in TBST for 2 hour at room temperature. The primary antibody (HY-P80599, 1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% defatted milk powder in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.

Immunocytochemistry analysis of NIH3T3 cells labeling CD31 with CD31 antibody (HY-P80599) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with CD31 antibody (HY-P80599) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Mouse IgG H&L(HY-P8005, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

Cell adhesion molecule which is required for leukocyte transendothelial migration (TEM) under most inflammatory conditions (PubMed:17580308, PubMed:19342684). Tyr-690 plays a critical role in TEM and is required for efficient trafficking of PECAM1 to and from the lateral border recycling compartment (LBRC) and is also essential for the LBRC membrane to be targeted around migrating leukocytes (PubMed:19342684). Trans-homophilic interaction may play a role in endothelial cell-cell adhesion via cell junctions (PubMed:27958302). Heterophilic interaction with CD177 plays a role in transendothelial migration of neutrophils (PubMed:17580308). Homophilic ligation of PECAM1 prevents macrophage-mediated phagocytosis of neighboring viable leukocytes by transmitting a detachment signal (PubMed:12110892). Promotes macrophage-mediated phagocytosis of apoptotic leukocytes by tethering them to the phagocytic cells; PECAM1-mediated detachment signal appears to be disabled in apoptotic leukocytes (PubMed:12110892). Tissue_specificity:This protein is expressed on platelets and leukocytes, primarily concentrated at the boundaries between endothelial cells (PubMed:18388311, PubMed:21464369) . It is also expressed (protein level) in human umbilical vein endothelial cells (HUVECs) (PubMed:17580308, PubMed:19342684) . It is also expressed (protein level) on neutrophils (PubMed:17580308) . The long subtype predominated in all tissues examined (PubMed:12433657) . The Δ12 subtype was detected only in the trachea (PubMed:12433657) . The Δ14-15 subtype was detected only in the lungs (PubMed:12433657) . The Delta14 subtype was detected in all tissues examined, with the strongest expression in the heart (PubMed:12433657) .
