Western blot analysis of extracts from Hela(lane 2(20ug) , Jurkat(lane 3(20ug) and C6(lane 4(20ug),3T3-L1(lane 5(20ug) using ATG7 Antibody (HY-P86509) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P83730, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

Immunocytochemistry analysis of HeLa cells labeling ATG7 with ATG7 Antibody (HY-P86509) at 1/200 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 in PBS for 15 minutes at room temperature, then blocked with quick block buffer for 10 minutes at room temperature. Cells were then incubated with ATG7 Antibody (HY-P86509) at 1/200 dilution in quick block buffer overnight at 4 ℃. AF488-conjugated Goat Anti-Rabbit IgG H&L(HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

Function
Atg7 is a core autophagy-related protein with E1-like enzyme activity, responsible for activating and transferring Atg8 (LC3/GABARAP) family proteins to facilitate autophagosome formation. In addition to its role in classical autophagy, Atg7 is involved in immune regulation (such as LC3-associated phagocytosis, LAP), cell cycle control, apoptosis, and non-conventional protein secretion. It plays a crucial role in maintaining organelle homeostasis, removing damaged cellular components, and responding to cellular stress. Loss of Atg7 can lead to neurodegenerative diseases such as Alzheimer’s disease, Parkinson’s disease, and amyotrophic lateral sclerosis. Moreover, Atg7 can both suppress tumor development and enhance tumor cell survival under extreme conditions. Atg7 also plays a significant role in infection defense and metabolic homeostasis, with its deficiency potentially increasing the risk of infections and being associated with metabolic diseases such as type 2 diabetes and fatty liver disease. Therefore, Atg7 is a potential research target for multiple diseases[1].
