Function
S100P is a member of the S100 family of small EF-hand calcium-binding proteins and functions as a mediator of Ca2+-dependent signal transduction, influencing cellular behavior through intracellular and extracellular mechanisms[1]. Mechanistically, extracellular S100P can bind the receptor for advanced glycation end products (RAGE), activating downstream signaling pathways including ERK and NF-κB that promote cell proliferation and survival[2]. Through these signaling networks, S100P has been implicated in the occurrence and progression of multiple human cancers and contributes to biological processes associated with tumor growth, migration, invasion, and metastatic potential[3][4]. In experimental cancer models, reduced S100P expression suppresses proliferative and invasive phenotypes, supporting its functional role in tumor progression[4][5]. Compared with related S100 family members that are predominantly encoded within the chromosome 1q21 S100 gene cluster, S100P is distinct because its gene is located on chromosome 4p16, indicating unique genomic organization within the family[7]. S100P also displays characteristic metal-binding properties, interacting not only with Ca2+ but also with Mg2+, Cu2+, and Zn2+, which may influence its structural and functional regulation[1]. For experimental applications, disruption of the S100P-RAGE interaction has been reported using compounds such as cromolyn and pentamidine, providing useful tools for mechanistic studies of S100P-driven signaling pathways[2].
