PRMT6 (protein arginine methyltransferase 6) is a type I protein arginine methyltransferase that catalyzes mono- and asymmetric dimethylation of arginine residues and functions predominantly as a nuclear epigenetic regulator of gene expression[1][8]. Mechanistically, PRMT6 deposits the repressive histone mark H3R2me2a, and this modification antagonizes H3K4 trimethylation, thereby influencing chromatin organization and transcriptional programs controlling cellular proliferation and differentiation[2][3]. Through regulation of H3R2 methylation at target promoters, PRMT6 modulates expression of cell-cycle regulators including p21 and p27, linking arginine methylation to G2 checkpoint control and cell-cycle progression in experimental models[4]. Beyond cell-cycle regulation, PRMT6 participates in broader epigenetic and DNA methylation networks through H3R2me2a-dependent effects on chromatin-associated regulatory machinery[8][5].
