Background

  • Function

TNFSF15 is a cytokine that transmits signals through the TNFRSF25 receptor. As a T-cell co-stimulatory molecule, it enhances IL-2 responsiveness and the secretion of pro-inflammatory cytokines, thereby promoting T-cell activation, proliferation, and survival of activated T cells. TNFSF15 inhibits vascular endothelial cell growth and angiogenesis. TNFSF15 binds to the decoy receptor TNFRSF6B, blocking activation that is normally triggered by TNFSF15, thereby reducing T cell activation and alleviating inflammation.

  • Subcellular Localization

Membrane; Single-pass type II membrane protein

  • Expression

Tissue specificity: Specifically expressed in endothelial cells. Detected in monocytes, placenta, lung, liver, kidney, skeletal muscle, pancreas, spleen, prostate, small intestine and colon.
Induction: Up-regulated by IL1A/interleukin-1 alpha and TNF.

  • Isoforms & Post-Translational Modification

    TNFSF15 has three isomers: TL1A; 28,087 Da (predicted); VEGI-192; 21,857 Da (predicted); VEGI-174; 20,131 Da (predicted).

  • Subunit

    Homotrimer. TNFSF15 binds TNFRSF6B with 3:3 stoichiometry.
