Western blot analysis of extracts from Jurkat(lane 2(20ug),HEK293T(lane 3(20ug) and Hela(lane 4(20ug) using Eg5 Antibody (HY-P80647) Mouse mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P83730, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

Immunocytochemistry analysis of HeLa cells labeling Eg5 Antibody with Eg5 Antibody (HY-P80647) at 1/100 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with BSA for Immunol Staining for 10 min at room temperature. Cells were then incubated with Eg5 Antibody (HY-P80647) at 1/100 dilution in BSA for Immunol Staining the night at 4 ℃. AF488-conjugated AffiniPure Goat Anti-Mouse IgG H&L(HY-P8005, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

Background

Function
Eg5 is a Motor protein required for establishing a bipolar spindle and thus contributing to chromosome congression during mitosis. Required in non-mitotic cells for transport of secretory proteins from the Golgi complex to the cell surface[1][2][3].

Subcellular Localization
Cytoplasm; Cytoplasm, cytoskeleton, spindle pole
-
Subunit
Interacts with the thyroid hormone receptor in the presence of thyroid hormone. Component of a large chromatin remodeling complex, at least composed of MYSM1, PCAF, RBM10 and KIF11/TRIP5. Interacts (via C-terminus) with the kinase NEK6 in both interphase and mitosis. Interacts with RARRES1 and AGBL2 (PubMed:21303978). Interacts with TPX2 (PubMed:37728657)
