Verification Images

  • Experimental Validation Results for MCM5 Antibody (YA9790)
Western blot analysis was performed on protein extracts (25 μg) from Ramos (lane 2), Jurkat (lane 3), U87 (lane 4), and 293T(lane 5) using MCM5 antibody. Proteins were transferred onto a 0.45 μm PVDF membrane using the Trans-Blot® Turbo system for 13 min. The membrane was then blocked with 5% nonfat milk in TBST (HY-K1025) for 1 h at room temperature. The primary antibody (1:1000) and loading control antibody GAPDH Antibody (HRP) (HY-P80954A) (1:5000) were diluted in 5% nonfat milk in TBST and incubated with the membrane overnight at 4°C. After washing, the membrane of primary antibody was incubated with HRP-conjugated goat anti-rabbit/mouse IgG secondary antibody (HY-P8001/HY-P8004) (1:5000) diluted in 5% nonfat milk in TBST for 1 h at room temperature. Protein bands were visualized using an Ultra High Sensitivity ECL detection kit (HY-K1005).

Background

  • Function

MCM5 acts as a component of the MCM2-7 complex (MCM complex) which is the replicative helicase essential for 'once per cell cycle' DNA replication initiation and elongation in eukaryotic cells. Core component of CDC45-MCM-GINS (CMG) helicase, the molecular machine that unwinds template DNA during replication, and around which the replisome is built. The active ATPase sites in the MCM2-7 ring are formed through the interaction surfaces of two neighboring subunits such that a critical structure of a conserved arginine finger motif is provided in trans relative to the ATP-binding site of the Walker A box of the adjacent subunit. The six ATPase active sites, however, are likely to contribute differentially to the complex helicase activity[1][2][3][4][5][6].

  • Subcellular Localization

    Nucleus; Chromosome

  • Isoforms & Post-Translational Modification

MCM5 has an amino acid length of 734, molecular weight is 82286 Da.
UFMylated at Lys-583; UFMylation occurs at the ongoing replication fork and promotes the CMG replicative DNA helicase complex formation.
