Verification Images

  • Experimental Validation Results for Growth hormone receptor Antibody
Western blot analysis of extracts from Raji (lane 2(20μg), Raji (lane 3(40μg), using Growth hormone receptor Antibody. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.

Background

  • Function

Growth hormone receptor receptor for pituitary gland growth hormone (GH1) involved in regulating postnatal body growth. On ligand binding, couples to the JAK2/STAT5 pathway; The soluble form (GHBP) acts as a reservoir of growth hormone in plasma and may be a modulator/inhibitor of GH signaling; Up-regulates the production of the soluble Growth hormone-binding protein form (GHBP) and acts as a negative inhibitor of growth hormone signaling[1][2][3][4].

  • Subcellular Localization

Cell membrane; Single-pass type I membrane protein; Cell membrane; Single-pass type I membrane protein; Secreted

  • Expression

Tissue_specificity:Expressed in various tissues with high expression in liver and skeletal muscle; Isoform 2 is expressed in lung, stomach and muscle; Predominantly expressed in kidney, bladder, adrenal gland and brain stem (PubMed:1569971) . Highly expressed in placental villi (PubMed:1569971, PubMed:8360189)

  • Isoforms & Post-Translational Modification

    P10912 has 4 isomers: P10912-1: 71500 Da (predicted); P10912-2: 34109 Da (predicted); P10912-3: 33889 Da (predicted); P10912-4: 69237 Da (predicted).
The soluble form (GHBP) is produced by phorbol ester-promoted proteolytic cleavage at the cell surface (shedding) by ADAM17/TACE (PubMed:11785980). Shedding is inhibited by growth hormone (GH) binding to the receptor probably due to a conformational change in GHR rendering the receptor inaccessible to ADAM17 (By similarity); On GH binding, phosphorylated on tyrosine residues in the cytoplasmic domain by JAK2;Ubiquitinated by the ECS(SOCS2) complex following ligand-binding and phosphorylation by JAK2, leading to its degradation by the proteasome (PubMed:21980433, PubMed:25505247, PubMed:31182716, PubMed:34857742). Regulation by the ECS(SOCS2) complex acts as a negative feedback loop of growth hormone receptor signaling (PubMed:21980433). Ubiquitination is not sufficient for GHR internalization (By similarity)
