Verification Images

  • Experimental Validation Results for Ubiquitin Conjugating Enzyme E2 C Antibody (YA2734)
Western blot analysis was performed on extracts from Hela (lane 1, 15 μg), MCF-7 (lane 2, 15 μg), 3T3 (lane 3, 15 μg), and COS-7 (lane 4, 15 μg) using Ubiquitin Conjugating Enzyme E2 C Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non - fat milk in TBST at 4°C overnight. The primary antibody (1:1000 dilution) and the loading control antibody (beta-Actin(HRP), HY-P80993, 1:5000 dilution) was incubated in 5% non-fat milk in TBST for 1 hour at 37°C. Goat Anti - Rabbit IgG - HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
  • Experimental Validation Results for Ubiquitin Conjugating Enzyme E2 C Antibody (YA2734)
Western blot analysis of extracts from HeLa (lane 2(20μg), HL-60 (lane 3(20μg), THP-1 (lane 4(20μg) and A549 (lane 5(20μg) using Ubiquitin Conjugating Enzyme E2 C Antibody (HY-P82989) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000)and Loading control antibody (Beta Actin, HY-P80993, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

Background

  • Function

Ubiquitin Conjugating Enzyme E2 C is an Accepts ubiquitin from the E1 complex and catalyzes its covalent attachment to other proteins. In vitro catalyzes 'Lys-11'- and 'Lys-48'-linked polyubiquitination. Acts as an essential factor of the anaphase promoting complex/cyclosome (APC/C), a cell cycle-regulated ubiquitin ligase that controls progression through mitosis. Acts by initiating 'Lys-11'-linked polyubiquitin chains on APC/C substrates, leading to the degradation of APC/C substrates by the proteasome and promoting mitotic exit

  • Isoforms & Post-Translational Modification

    O00762 has 4 isomers: O00762-1: 19652 Da (predicted); O00762-2: 15765 Da (predicted); O00762-3: 16676 Da (predicted); O00762-4: 17373 Da (predicted).
Autoubiquitinated by the APC/C complex, leading to its degradation by the proteasome. Its degradation plays a central role in APC/C regulation, allowing cyclin-A accumulation before S phase entry. APC/C substrates inhibit the autoubiquitination of UBE2C/UBCH10 but not its E2 function, hence APC/C remaining active until its substrates have been destroyed

  • Subunit

Component of the APC/C complex, composed of at least 14 distinct subunits that assemble into a complex of at least 19 chains with a combined molecular mass of around 1.2 MDa. Within this complex, directly interacts with ANAPC2
