  • Experimental Validation Results for Cyclin A1 Antibody
Western blot analysis of extracts from NIH/3T3(lane 2(20ug) ,K562(lane 3(20ug) and Jurkat(lane 4(20ug) using Cyclin A1 Antibody (HY-P80631) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
  • Experimental Validation Results for Cyclin A1 Antibody
Immunocytochemistry analysis of Hela cells labeling Cyclin A1 with Cyclin A1 Antibody (HY-P80631) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with Cyclin A1 Antibody (HY-P80631) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L(HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

  • Experimental Validation Results for Cyclin A1 Antibody
Immunohistochemical analysis of paraffin-embedded Mouse brain tissue using Cyclin A1 Antibody. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 2 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P80631, 1/100) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background

  • Function

Cyclin A1 may be involved in the control of the cell cycle at the G1/S (start) and G2/M (mitosis) transitions. May primarily function in the control of the germline meiotic cell cycle and additionally in the control of mitotic cell cycle in some somatic cells

Tissue_specificity:Very high levels in testis and very low levels in brain. Also found in myeloid leukemia cell lines

  • Isoforms & Post-Translational Modification

    P78396 has 3 isomers: P78396-1: 52358 Da (predicted); P78396-2: 52230 Da (predicted); P78396-3: 47494 Da (predicted).
Polyubiquitinated via 'Lys-11'-linked ubiquitin by the anaphase-promoting complex (APC/C), leading to its degradation by the proteasome (PubMed:21596315). Deubiquitinated and stabilized by USP37 enables entry into S phase (PubMed:21596315). Ubiquitinated during the G1 phase by the SCF(FBXO31) complex, leading to its proteasomal degradation (PubMed:31413110)

  • Subunit

Interacts with the CDK2 and the CDC2 protein kinases to form a serine/threonine kinase holoenzyme complex. The cyclin subunit imparts substrate specificity to the complex. Does not bind CDK4 and CDK5 (in vitro). The cyclin A1-CDK2 complex interacts with transcription factor E2F-1 and RB proteins. Found in a complex with CDK2, CABLES1 and CCNE1 (By similarity). Interacts with INCA1 (PubMed:15159402, PubMed:21540187). Interacts with KLHDC9 (PubMed:15159402)
