  • Western blot analysis of extracts from HeLa (lane 1(20μg)) 、HepG2 (lane 2(20μg)) 、A549 (lane 3(20μg)) 、C2C12 (lane 4(20μg)) 、C6 (lane 5(20μg)) 、HEK293 (lane 6(20μg)) 、Mouse kidney (lane 7(20μg)) 、Rat kidney (lane 8(20μg)) and Mouse spleen (lane 9(20μg)) using Bax Antibody (HY-P80027) . Proteins were transferred to a PVDF membrane and blocked with 5% nonfat dry milk in TBST for 1.5 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Hsp90, 1/10000) was used in 5% nonfat dry milk in TBST at 4℃ overnight. Goat Anti-Mouse IgG-HRP Secondary Antibody (1/10,000) was used for 1 hour at room temperature.

  • Western blot analysis of extracts from Hela (lane2(20μg), MCF-7 (lane3(20μg), C2C12 (lane4(20μg) and C6 (lane5(20μg) using Bax Antibody (HY-P80027). Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80993, 1/10,000) was used in 5% non-fat milk in TBST for 2 hour at room temperature. Goat Anti-Mouse IgG-HRP Secondary Antibody (HY-P8004, 1/10,000) was used for 1 hour at room temperature.

  • Immunohistochemical analysis of paraffin-embedded Rat kidney tissue using BAX Antibody (YA825). The section was pre-treated using heat mediated antigen retrieval with Tris-EDTA buffer (pH 9.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (HY-P80027, 1/100) in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Function:Plays a role in the mitochondrial apoptotic process (PubMed:10772918, PubMed:11060313, PubMed:16113678, PubMed:16199525, PubMed:18948948, PubMed:21199865, PubMed:21458670, PubMed:25609812, PubMed:36361894, PubMed:8358790, PubMed:8521816). Under normal conditions, BAX is largely cytosolic via constant retrotranslocation from mitochondria to the cytosol mediated by BCL2L1/Bcl-xL, which avoids accumulation of toxic BAX levels at the mitochondrial outer membrane (MOM) (PubMed:21458670). Under stress conditions, undergoes a conformation change that causes translocation to the mitochondrion membrane, leading to the release of cytochrome c that then triggers apoptosis (PubMed:10772918, PubMed:11060313, PubMed:16113678, PubMed:16199525, PubMed:18948948, PubMed:21199865, PubMed:21458670, PubMed:25609812, PubMed:8358790, PubMed:8521816). Tissue_specificity:This gene is expressed in a variety of tissues. The Psi isoform is present in gliomas. The α isoform is expressed in the spleen, breast, ovary, testis, colon, and brain, and at low levels in the skin and lungs. The Sigma isoform is expressed in the spleen, breast, ovary, testis, lung, colon, and brain, and at low levels in the skin.
