  • Western blot analysis was performed on extracts from Hela (lane 1, 15 μg), Jurkat (lane 2, 15 μg), A549 (lane 3, 15 μg), and MDA-MB-231 (lane 4, 15 μg) using LYRIC/AEG1 Mouse mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight. The primary antibody (1:1000 dilution) and the loading control antibody (beta-Actin, HY-P80438, 1:5000 dilution) was incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Mouse IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.

  • Immunohistochemical analysis of paraffin-embedded human Breast Cancer tissue using LYRIC antibody was performed. The section was pretreated using high-temperature and high-pressure mediated EDTA antigen retrieval buffer (pH 9.0), for 5 minutes. The tissues were incubated with primary antibody (HY-P81286, 1:50 dilution) at room temperature for 60 minutes. Detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. The tissues were counterstained with hematoxylin and mounted with neutral balsam mounting medium.

  • Tyramide signaling amplification based immunofluorescence was performed on paraffin-embedded human Colon cancer tissue using LYRIC antibody. Antigen retrieval was performed in EDTA buffer pH 9.0 (95 °C, 20 min) followed by cooling to RT. Then incubated with primary antibody (HY-P81286, 1:50 dilution) at room temperature for 60 minutes and HRP conjugated secondary antibody for 10 minutes. Fluorescence was then developed with Vari Fluor 532 TSA (200×)(HY-D1832). The tissues were counterstained with DAPI and mounted with Anti-fade mounting medium.

Background
Function:Down-regulates SLC1A2/EAAT2 promoter activity when expressed ectopically. Activates the nuclear factor kappa-B (NF-kappa-B) transcription factor. Promotes anchorage-independent growth of immortalized melanocytes and astrocytes which is a key component in tumor cell expansion. Promotes lung metastasis and also has an effect on bone and brain metastasis, possibly by enhancing the seeding of tumor cells to the target organ endothelium. Induces chemoresistance
Subcellular Localization:Endoplasmic reticulum membrane; Single-pass membrane protein; Nucleus membrane; Single-pass membrane protein; Cell junction, tight junction; Nucleus, nucleolus; Cytoplasm, perinuclear region
Expression:
Tissue_specificity:This gene is widely expressed in muscle-based organs (such as skeletal muscle, heart, tongue, and small intestine) and endocrine glands (such as the thyroid and adrenal glands) , with the highest expression levels. It is overexpressed in a variety of cancers, including breast cancer, brain cancer, prostate cancer, melanoma, and glioblastoma multiforme.

Induction:By TNF (at protein level) . By HIV-1 infection of primary fetal astrocytes
Subunit:Interacts with BCCIP, CREBBP/CBP and RELA/p65

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
