Western blot analysis was performed on extracts from Recombinant CBP-Tag protein-10ng (lane 1, 10 ng), Recombinant CBP-Tag protein-5ng (lane 2, 5 ng), Recombinant CBP-Tag protein-2ng (lane 3, 2 ng), and Recombinant CBP-Tag protein-1ng (lane 4, 1 ng) using CBP Tag Mouse mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non - fat milk in TBST at 4°C overnight. The primary antibody (1:1000 dilution) was incubated in 5% non-fat milk in TBST for 1 hour at 37°C. Goat Anti - Mouse IgG - HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.

CBP Tag is a CBP affinity tag is a useful tag in protein study which can be added to the N or C terminus of proteins of interest through DNA recombinant technology. The tag is derived from muscle myosin light-chain kinase. The tag comprises 26 amino acid residues with the molecular weight of 4 kDa and sequence of KRRWKKNFIAVSAANRFKKISSSGAL. CBP tag has the relatively high affinity for calmodulin (CaM).
