    Western blot analysis of extracts from Human brain cancer (lane 1(20μg)) 、Mouse brain (lane 2(20μg)) 、unboiled Mouse brain (lane 3(20μg)) 、Rat brain (lane 4(20μg)) and unboiled Rat brain (lane 5(20μg)) using NMDAR1 Antibody (HY-P87676) . Proteins were transferred to a PVDF membrane and blocked with 5% nonfat dry milk in TBST for 1.5 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (β Actin, 1/10000) was used in 5% nonfat dry milk in TBST at 4℃ overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (1/10,000) was used for 1 hour at room temperature.

Background

  • Function

    NMDAR1 component of N-methyl-D-aspartate (NMDA) receptors (NMDARs) that function as heterotetrameric, ligand-gated cation channels with high calcium permeability and voltage-dependent block by Mg(2+). NMDARs participate in synaptic plasticity for learning and memory formation by contributing to the long-term potentiation (LTP). Channel activation requires binding of the neurotransmitter L-glutamate to the GluN2 subunit, glycine or D-serine binding to the GluN1 subunit, plus membrane depolarization to eliminate channel inhibition by Mg(2+). NMDARs mediate simultaneously the potasium efflux and the influx of calcium and sodium. Each GluN2 or GluN3 subunit confers differential attributes to channel properties, including activation, deactivation and desensitization kinetics, pH sensitivity, Ca2(+) permeability, and binding to allosteric modulators[1][2][3][4][5][6][7][8][9][10][11][12][13][14][15].
