Western blot analysis of extracts from A549 (lane 2(20μg), K562 (lane 3(20μg), 3T3 (lane 4(20μg) and Hela (lane 5(20μg) using Nogo B Receptor Antibody (HY-P81669) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000)and Loading control antibody (Beta Actin, HY-P80993, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

Function
Nogo B Receptor is a With DHDDS, forms the dehydrodolichyl diphosphate synthase (DDS) complex, an essential component of the dolichol monophosphate (Dol-P) biosynthetic machinery. Both subunits contribute to enzymatic activity, i.e. condensation of multiple copies of isopentenyl pyrophosphate (IPP) to farnesyl pyrophosphate (FPP) to produce dehydrodolichyl diphosphate (Dedol-PP), a precursor of dolichol phosphate which is utilized as a sugar carrier in protein glycosylation in the endoplasmic reticulum (ER). Synthesizes long-chain polyprenols, mostly of C95 and C100 chain length. Regulates the glycosylation and stability of nascent NPC2, thereby promoting trafficking of LDL-derived cholesterol. Acts as a specific receptor for the N-terminus of Nogo-B, a neural and cardiovascular regulator[1][2][3][4][5][6].

Subunit
The active dehydrodolichyl diphosphate synthase complex is a heterotetramer composed of a dimer of heterodimer of DHDDS and NUS1 (PubMed:19723497, PubMed:25066056, PubMed:28842490, PubMed:32817466, PubMed:33077723, PubMed:35584224). Interacts with NPC2 (PubMed:21572394)
