Flow cytometric analysis of 1X10^6 Hela cells labeling BMPR1A Antibody(red). Cells were fixed with 4% paraformaldehyde and permeabilised with 0.2% Triton X-100. Then stained with the primary antibody at 1/400 dilution overnight at 4℃. AF 488 Goat Anti-mouse IgG H&L was used as the secondary antibody at 1/1,000 dilution for 45 minutes at room temperature. Mouse IgG Isotype Control (blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).

Immunofluorescence analysis of HepG2 cells labeling BMPR1A antibody at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 in PBS for 15 minutes at room temperature, then blocked with 1% BSA in 10% negative goat serum for 1 hour at room temperature.Cells were then incubated with BMPR1A antibody at 1/50 dilution in 1% BSA in PBST overnight at 4 ℃. AF 488 Goat Anti-mouse IgG H&L (green) was used as the secondary antibody at 1/500 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue)
Background

Function
BMPR1A is an On ligand binding, forms a receptor complex consisting of two type II and two type I transmembrane serine/threonine kinases. Type II receptors phosphorylate and activate type I receptors which autophosphorylate, then bind and activate SMAD transcriptional regulators. Receptor for BMP2, BMP4, GDF5 and GDF6. Positively regulates chondrocyte differentiation through GDF5 interaction. Mediates induction of adipogenesis by GDF6. May promote the expression of HAMP, potentially via its interaction with BMP2

Subcellular Localization
Cell membrane; Single-pass type I membrane protein; Cell surface

Expression
Tissue_specificity:Highly expressed in skeletal muscle -
Subunit
Interacts with low affinity with GDF5; positively regulates chondrocyte differentiation (PubMed:24098149). Interacts with BMP4 (PubMed:8006002). Interacts with SCUBE3 (PubMed:33308444). Interacts with TSC22D1/TSC-22 (PubMed:21791611). Interacts with BMP2; the interaction may induce HAMP expression (PubMed:10881198, PubMed:15064755, PubMed:17295905, PubMed:18937504, PubMed:31800957). Interacts with BMP6 (PubMed:31800957). Interacts with heterodimers composed of BMP2 and BMP6 in vitro; the interaction may induce HAMP expression (PubMed:31800957). Interacts with TGFBR3 (PubMed:19726563)
