    Western blot analysis of extracts from MCF-7 (lane 2(20μg) and MCF-7 (lane 3(40μg) using CD42a (HY-P80059) Mouse mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

    Immunohistochemical analysis of paraffin-embedded mouse spleen tissue using CD42a Antibody. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 8 minutes. The tissues were blocked in QuickBlock for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody at 1/100 dilution in 4℃ overnight. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background

  • Function

    CD42a is a platelet membrane glycoprotein encoded by the GP9 gene and is an important component of the GPIb-IX-V complex. CD42a is mainly expressed in platelets and megakaryocytes. Although it does not directly bind to vWF, it is crucial for the stable expression and normal function of the complex. CD42a participates in the initial adhesion of platelets to damaged blood vessel walls and is a key early molecule in the hemostatic response. CD42a is an important pathogenic protein associated with Bernard-Soulier syndrome and is also a commonly used biomarker in megakaryocyte differentiation, AML-M7 immunophenotyping, and platelet function testing.
