Western blot analysis was performed on extracts from Jurkat (lane 1, 15 μg), MDA-MB-231 (lane 2, 15 μg), Hela (lane 3, 15 μg), and U-87MG (lane 4, 15 μg) using Lamin B2 Mouse mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight.The primary antibody (1:1000 dilution) and the loading control antibody (beta-Tubulin, HY-P80955, 1:5000 dilution) was incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Mouse IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.

Lamin B2 is a Lamins are intermediate filament proteins that assemble into a filamentous meshwork, and which constitute the major components of the nuclear lamina, a fibrous layer on the nucleoplasmic side of the inner nuclear membrane. Lamins provide a framework for the nuclear envelope, bridging the nuclear envelope and chromatin, thereby playing an important role in nuclear assembly, chromatin organization, nuclear membrane and telomere dynamics. The structural integrity of the lamina is strictly controlled by the cell cycle, as seen by the disintegration and formation of the nuclear envelope in prophase and telophase, respectively[1].
