Western blot analysis of extracts from Jurkat(lane 2(20ug) , NIH/3T3(lane 3(20ug) and HEK293(lane 4(20ug) using GADD45A Antibody (HY-P81331) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.
Background
-
Function
GADD45A is an In T-cells, functions as a regulator of p38 MAPKs by inhibiting p88 phosphorylation and activity. Might affect PCNA interaction with some CDK (cell division protein kinase) complexes; stimulates DNA excision repair in vitro and inhibits entry of cells into S phase

Subcellular Localization
Nucleus
-
Expression
Induction:By UV irradiation, X-rays, growth arrest and alkylating agents. The induction is mediated by some kinase (s) other than PKC -
Isoforms & Post-Translational Modification
P24522 has 2 isomers: P24522-1: 18336 Da (predicted); P24522-2: 14622 Da (predicted).
-
Subunit
Interacts with MAPK14 (By similarity). Predominantly monomeric but also forms dimers and other oligomers as concentration increases. Interacts with GADD45GIP1. Interacts weakly with PCNA. Interacts with AURKA, likely to compete with dimerization
