  • Experimental Validation Results for ENT2 Antibody (YA2442)
    Western blot analysis of extracts from Hela (lane2, 20μg), K562 (lane3, 20μg) and Jurkat (lane4, 20μg) using ENT2 Antibody (HY-P82697). Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80993, 1/10,000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (HY-P8001,1/10,000) was used for 1 hour at room temperature.

    Equilibrative nucleoside transporter 2 (ENT2, SLC29A2) is a broadly expressed equilibrative transporter that mediates bidirectional flux of purine and pyrimidine nucleosides as well as multiple nucleobases, thereby supporting nucleoside homeostasis and salvage pathways of nucleotide synthesis[1][2]. ENT2 functions as a sodium-independent membrane transporter and contributes to the regulation of extracellular and intracellular pools of adenosine, inosine, hypoxanthine, and related metabolites that influence cellular metabolism and signaling[1]. Mechanistically, ENT2 participates in nucleoside transport processes that affect adenosine receptor-dependent signaling and tissue responses to metabolic stress[1][3]. In disease-relevant settings, ENT2 has been implicated in intestinal epithelial adenosine regulation, where repression or deletion of ENT2 enhances mucosal A2B receptor signaling associated with resolution of intestinal inflammation[3]. ENT2 also contributes to the cellular disposition of therapeutic nucleoside analogs, including anticancer and antiviral agents, making transporter activity an important determinant of drug uptake and pharmacological response[1][2]. Compared with the closely related isoform ENT1, ENT2 shares broad nucleoside selectivity but displays efficient transport of nucleobases and is characteristically insensitive to nitrobenzylmercaptopurine riboside (NBMPR), a distinction widely used for functional discrimination in transport studies[1][2]. For experimental applications, NBMPR sensitivity profiles, together with dipyridamole and dilazep inhibition studies, are commonly used to distinguish ENT family transporter activity and to investigate nucleoside transport mechanisms in cellular models[1].
