Verification Images

  • Experimental Validation Results for Arginase-1 Antibody (YA3647)
Western blot analysis of extracts from Rat liver tissue (lane2, 20μg), HepG2 (lane3, 20μg), HuH-7 (lane4, 20μg), Rat liver tissue (lane5, 30μg) and HepG2 (lane6, 30μg) using Arginase-1 Antibody (HY-P83950). Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80993, 1/10,000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse IgG-HRP Secondary Antibody (HY-P8004,1/10,000) was used for 1 hour at room temperature.
  • Experimental Validation Results for Arginase-1 Antibody (YA3647)
Flow cytometric analysis of 1X106 HepG2 cells labeling Arginase-1 Antibody (HY-P83950, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/400 dilution for an hour at 4℃. AF488-conjugated Goat Anti-Mouse IgG H&L (HY-P8005) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Mouse IgG Isotype Control (HY-P80757, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).

Background

  • Function

Arginase-1 is key element of the urea cycle converting L-arginine to urea and L-ornithine, which is further metabolized into metabolites proline and polyamides that drive collagen synthesis and bioenergetic pathways critical for cell proliferation, respectively; the urea cycle takes place primarily in the liver and, to a lesser extent, in the kidneys; Functions in L-arginine homeostasis in nonhepatic tissues characterized by the competition between nitric oxide synthase (NOS) and arginase for the available intracellular substrate arginine. Arginine metabolism is a critical regulator of innate and adaptive immune responses. Involved in an antimicrobial effector pathway in polymorphonuclear granulocytes (PMN). Upon PMN cell death is liberated from the phagolysosome and depletes arginine in the microenvironment leading to suppressed T cell and natural killer (NK) cell proliferation and cytokine secretion. In group 2 innate lymphoid cells (ILC2s) promotes acute type 2 inflammation in the lung and is involved in optimal ILC2 proliferation but not survival. In humans, the immunological role in the monocytic/macrophage/dendritic cell (DC) lineage is unsure[1][2][3].

  • Expression

Tissue_specificity:In the immune system, it was initially reported to be selectively expressed in granulocytes (polymorphonuclear leukocytes [PMN]) (PubMed:15546957) . It has also been detected in macrophages of mycobacterial granulomas (PubMed:23749634) . During lung disease, it is expressed in type 2 innate lymphocytes (ILC2) (PubMed:27043409) .

    Induction:By arginine or homoarginine
