Western blot analysis of extracts from HEK293T (lane 2(20μg), HepG2 (lane 3(20μg), SH-SY5Y (lane 4(20μg), HCT116 (lane 5(20μg) using PHD2 Antibody. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody (1/1000)and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.

Function
PHD2 is a HIF prolyl hydroxylase that regulates HIF-1 protein levels in mammalian cells. HIF-1α prolyl hydroxylases mainly include PHD1, PHD2, and PHD3. These three enzymes have sequence-similar catalytic domains and conserve amino acid residues for coordinating Fe (II) ions. Under hypoxic conditions, due to insufficient oxygen supply, the hydroxylase activity of PHDs decreases, leading to HIF-1α accumulation and inducing the expression of HRE-regulated target genes. Therefore, PHDs are considered intracellular oxygen sensors. PHD1 is found only in the cell nucleus, while PHD2 is mainly located in the cytoplasm.
