Background

  • Function

NFAT5 transcription factor involved, among others, in the transcriptional regulation of osmoprotective and inflammatory genes. Binds the DNA consensus sequence 5'-[ACT][AG]TGGAAA[CAT]A[TA][ATC][CA][ATG][GT][GAC][CG][CT]-3'. Mediates the transcriptional response to hypertonicity. Positively regulates the transcription of LCN2 and S100A4 genes; optimal transactivation of these genes requires the presence of DDX5/DDX17. Also involved in the DNA damage response by preventing formation of R-loops; R-loops are composed of a DNA:RNA hybrid and the associated non-template single-stranded DNA[1][2][3][4].

  • Subcellular Localization

    Nucleus; Cytoplasm; Chromosome

  • Expression

Tissue_specificity:Widely expressed, with highest levels in skeletal muscle, brain, heart and peripheral blood leukocytes

    O94916 has 5 isomers: O94916-1: 165763 Da (predicted); O94916-2: 157983 Da (predicted); O94916-3: 10459 Da (predicted); O94916-4: 167737 Da (predicted); O94916-5: 167808 Da (predicted).
Phosphorylated (PubMed:10377394). Phosphorylated at Thr-135 by CDK5 in response to osmotic stress; this phosphorylation mediates its rapid nuclear localization (PubMed:21209322);Poly-ADP-ribosylated by PARP1 in response to DNA damage, promoting recruitment to sites of R-loop-associated DNA damage

  • Subunit

Homodimer when bound to DNA, completely encircles its DNA target (PubMed:11780147). Interacts with CIDEC; this interaction is direct and retains NFAT5 in the cytoplasm (By similarity). Does not bind with Fos and Jun transcription factors (PubMed:10377394). Interacts with DDX5 and DDX17; this interaction leads to DDX5/DDX17 recruitment to LNC2 and S100A4 promoters and NFAT5-mediated DDX5/DDX17-enhanced transactivation (PubMed:22266867)
