Western blot analysis of extracts from K562 (lane 2(20μg), 3T3 (lane 3(20μg), Hela (lane 4(20μg) using Glutaminase Antibody. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody and Loading control antibody (Beta Actin, HY-P80438, 1/3000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (HY-P8004/HY-P8001, 1/10,000) was used for 1 hour at room temperature.
Immunocytochemistry analysis of Siha cells labeling Glutaminase with Glutaminase Antibody (HY-P80690) at 1/50 dilution. Cells were fixed in 4% paraformaldehyde for 15 minutes at room temperature, permeabilized with 0.1% Triton X-100 for 10 minutes at room temperature, then blocked with QuickBlock™ Blocking Buffer for Immunol Staining for 10 min at room temperature. Cells were then incubated with Glutaminase Antibody (HY-P80690) at 1/50 dilution in QuickBlock™ Blocking Buffer for Immunol Staining at 4 ℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L（HY-P8002, Green) was used as the secondary antibody at 1/1,000 dilution. PBS instead of the primary antibody was used as the secondary antibody only control. The Nuclear counterstain was DAPI (Blue).

Background

Function：Catalyzes the first reaction in the primary pathway for the renal catabolism of glutamine. Plays a role in maintaining acid-base homeostasis. Regulates the levels of the neurotransmitter glutamate, the main excitatory neurotransmitter in the brain (PubMed:30239721, PubMed:30575854, PubMed:30970188); Lacks catalytic activity

Expression：
Tissue_specificity:Isomers 1 and 3 were detected in the cerebral cortex. Isomer 3 was highly expressed in astrocytomas, gangliogliomas, and ependymomas. Isomer 1 was highly expressed in the brain and kidneys, but not detected in the liver. Isomer 3 was highly expressed in the heart and pancreas, and expressed at lower levels in the placenta, lungs, pancreas, and kidneys, but not detected in the liver. Isomer 2 was expressed in cardiac and skeletal muscle.

Isoforms & Post-Translational Modification：O94925 has 3 isomers: O94925-1: 73461 Da (predicted); O94925-2: 17738 Da (predicted); O94925-3: 65460 Da (predicted).

The tetramers can assemble into rod-like oligomers (in vitro), but the physiological significance of this is not clear (By similarity). Interacts with RAF1 and MAP2K2 (PubMed:22538822). Interacts with ATCAY; the interaction is direct and may control GLS localization, negatively regulating its activity
