Western blot analysis of extracts from HepG2 (lane2(20μg), Hela (lane3(20μg), HL-60 (lane4(20μg) and Ramos (lane5(20μg) using CD322 Antibody (HY-P84414). Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80993, 1/10,000) was used in 5% non-fat milk in TBST for 2 hour at room temperature. Goat Anti-Mouse IgG-HRP Secondary Antibody (HY-P8004, 1/10,000) was used for 1 hour at room temperature.

Flow cytometric analysis of 1X106 HeLa cells labeling CD322 Antibody (HY-P84414, red). Cells were fixed with 4% paraformaldehyde. Then stained with the primary antibody at 1/50 dilution for an hour at 4℃. AF 488-conjugated AffiniPure Goat Anti- Mouse IgG H&L (HY-P8005) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Mouse IgG Isotype Control (HY-P80757, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).
Background
-
Function
CD322 regulates a variety of cellular processes by mediating interactions between heterologous cells through its homologous receptor JAM3. CD322 plays a role in the homing and mobilization of hematopoietic stem cells and progenitors in the bone marrow. On the surface of bone marrow stromal cells, CD322 facilitates the retention of hematopoietic stem cells and progenitors expressing JAM3. CD322 plays a central role in leukocyte exudation, promoting not only transendothelial migration of leukocytes but also adhesion and rolling of leukocytes along endothelial cells. Leukocyte adhesion and rolling depend on the binding of integrin α4/β1 to CD322[1].

Expression
Tissue_specificity:Highly expressed in heart, placenta, lung, foreskin and lymph node (PubMed:10779521, PubMed:10945976) . Prominently expressed on high endothelial venules and also present on the endothelia of other vessels (at protein level) (PubMed:10779521, PubMed:10945976) . Also expressed in the brain in the caudate nuclei (PubMed:31851307) -
