  • Experimental Validation Results for NCF1 Antibody (YA2575)
Western blot analysis was performed on extracts from Raji (lane 1, 15 μg), Ramos (lane 2, 15 μg), and Daudi (lane 3, 15 μg) using NCF1 Rabbit mAb.Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST at 4°C overnight. The primary antibody (1:1000 dilution) and the loading control antibody (GAPDH, HY-P80137, 1:60000 dilution) were incubated in 5% non-fat milk in TBST for 1 hour at 37°C.Goat Anti-Rabbit IgG-HRP Secondary Antibody (1:20000 dilution) was then applied for 40 minutes at 37°C.
  • Experimental Validation Results for NCF1 Antibody (YA2575)
Western blot analysis of extracts from Raji(lane 2(20ug) and Raji(lane 3(40ug) using NCF1 Antibody (HY-P82830) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P83730, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

Background

  • Function

NCF1 is a Subunit of the phagocyte NADPH oxidase complex that mediates the transfer of electrons from cytosolic NADPH to O2 to produce the superoxide anion (O2(-)). In the activated complex, electrons are first transferred from NADPH to flavin adenine dinucleotide (FAD) and subsequently transferred via two heme molecules to molecular oxygen, producing superoxide through an outer-sphere reaction. Activation of the NADPH oxidase complex is initiated by the assembly of cytosolic subunits of the NADPH oxidase complex with the core NADPH oxidase complex to form a complex at the plasma membrane or phagosomal membrane. This activation process is initiated by phosphorylation dependent binding of the cytosolic NCF1/p47-phox subunit to the C-terminus of CYBA/p22-phox[1][2][3][4][5].

  • Subcellular Localization

  • Expression

Tissue_specificity:Detected in peripheral blood monocytes and neutrophils (at protein level)

  • Isoforms & Post-Translational Modification

    P14598 has 2 isomers: P14598-1: 44682 Da (predicted); P14598-2: 30811 Da (predicted).
Phosphorylated by PRKCD; phosphorylation induces activation of NCF1, leading to assembly and activation of the NADPH oxidase complex

  • Subunit

Component of the phagocyte NADPH oxidase complex composed of an obligatory core heterodimer formed by the membrane proteins CYBA and CYBB and the cytosolic regulatory subunits NCF1/p47-phox, NCF2/p67-phox, NCF4/p40-phox and the small GTPase RAC1 or RAC2 (PubMed:38355798). Part of a cytosolic complex composed at least by NCF1, NCF2 and NCF4 (PubMed:8280052). Interacts (via C-terminus) with NCF2 (via the C-terminal SH3 domain) (PubMed:12169629). Interacts with NCF4 (PubMed:15657040). Interacts with CYBB (PubMed:9224653). Interacts (via the second SH3 domain) with CYBA; interaction is phosphorylation-dependent (PubMed:12732142, PubMed:16326715). Interacts with NOXA1 (PubMed:12716910). Interacts with ADAM15 (PubMed:19718658). Interacts with TRAF4 (PubMed:12023963, PubMed:16052631). Interacts with FASLG (PubMed:19807924). Interacts with PARK7 (via C-terminus); the interaction is enhanced by LPS and modulates NCF1 phosphorylation and membrane translocation (By similarity)
