    Western blot analysis of extracts from Hela(lane 2(20ug) , K562(lane 3(20ug)and Jurkat(lane 4(20ug) using ABL2 Antibody (HY-P83458) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

    Flow cytometric analysis of 1X10^6 Raji cells labeling ABL2 Antibody (HY-P83458, red). Cells were fixed with 4% paraformaldehyde and permeabilised with 90% methanol. Then stained with the primary antibody at 1/100 dilution for an hour at 4℃. Alexa Fluor® 488-conjugated AffiniPure Goat Anti-Rabbit IgG H&L (HY-P8002) was used as the secondary antibody at 1/1,000 dilution for 30 minutes at 4℃. Rabbit IgG Isotype Control (HY-P80879, blue) was used as the isotype control, cells without incubation with primary antibody were used as the unlabeled control (black).

    ABL2 is a non-receptor tyrosine protein kinase that plays overlapping roles with ABL1 in key processes of cell growth and survival, such as cytoskeleton remodeling in response to extracellular stimuli, cell movement and adhesion, and receptor endocytosis. ABL2 coordinates actin remodeling by tyrosine phosphorylation of proteins that control cytoskeleton dynamics, such as MYH10 (involved in movement), CTTN (involved in signal transduction), or TUBA1 and TUBB (microtubule subunits). It directly binds to F-actin and regulates actin cytoskeleton structure through its F-actin bundle activity. ABL2 participates in the regulation of cell adhesion and movement by phosphorylating key regulators such as CRK, CRKL, DOK1, or ARHGAP35. Adhesion-dependent phosphorylation of ARHGAP35 promotes its binding to RASA1, thereby recruiting ARHGAP35 to the cell periphery, where it inhibits RHO. ABL2 phosphorylates multiple receptor tyrosine kinases, such as PDGFRB, and other substrates involved in the regulation of endocytosis, such as RIN1.
