Background

Function
GLI2 is a Transcription factor that acts as a key effector of the smoothened signaling pathway, and which plays a role in skeleton development. Specifically binds to the minimal GLI-consensus sequence 5'-GACCACCCA-3' to activate or repress the expression of target genes (By similarity). Functions both as a transcriptional activator and repressor: the full-length GLI2 form (Transcription activator GLI2) acts as a transcription activator following smoothened activation, while the Transcription repressor GLI2R, which is generated in absence of smoothened, acts as a transcription repressor (By similarity). The transcriptional activator form constitutes the major form of GLI2 (By similarity)[1][2][3][4][5][6][7][8].

Expression
Tissue_Specificity: Expressed in breast cancers (at protein level). Isoform 1 and isoform 4 are expressed in HTLV-1-infected T-cell lines (at protein level). Isoform 1 and isoform 2 are strongly expressed in HTLV-1-infected T-cell lines. Isoform 3 and isoform 4 are weakly expressed in HTLV-1-infected T-cell lines. -
Isoforms & Post-Translational Modification
GLI2 has 5 isoforms, P10070-5: amino acid length is 1586, molecular weight is 167783 Da (predicted); P10070-1: amino acid length is 1258, molecular weight is 133241 Da (predicted); P10070-2: amino acid length is 1241, molecular weight is 131659 Da (predicted); P10070-3: amino acid length is 829, molecular weight is 88065 Da (predicted); P10070-4: amino acid length is 812, molecular weight is 86482 Da (predicted).
Acetylation at Lys-757 inhibits Hh target gene expression, probably by impeding entry into chromatin thus preventing promoter occupancy. -
Subunit
Interacts with SUFU; sequestering GLI2 in the cytoplasm.
