Western blot analysis of extracts from RAW264.7 (lane 2, 20ug), RAW264.7 treated with 5μg/mL LPS for 8 hours (lane3, 20μg) and RAW264.7 treated with 10μg/mL LPS for 8 hours (lane4, 20μg) using Phospho-ELK1 (Ser383) Antibody (HY-P810618). Proteins were transferred to a PVDF membrane and blocked with 5% BSA in TBST for 2 hour at room temperature. The primary antibody (1/5000) and Loading control antibody (Beta Actin, HY-P80993, 1/10,000) was used in 5% BSA in TBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (HY-P8001,1/10,000) was used for 1 hour at room temperature.

ELK1 is a Transcription factor that binds to purine-rich DNA sequences. Forms a ternary complex with SRF and the ETS and SRF motifs of the serum response element (SRE) on the promoter region of immediate early genes such as FOS and IER2. Induces target gene transcription upon JNK and MAPK-signaling pathways stimulation[1][2][3].
