  • Experimental Validation Results for CD38 Antibody (YA6174)
    Western blot analysis was performed on protein extracts (25 μg) from Mouse spleen (lane 2) and Mouse colon (lane 3) using CD38 antibody. Proteins were transferred onto a 0.45 μm PVDF membrane using the Trans-Blot® Turbo system for 13 min. The membrane was then blocked with 5% nonfat milk in TBST (HY-K1025) for 1 h at room temperature. Thhe primary antibody (1:1000) and loading control antibody GAPDH Antibody (HRP) (HY-P80954A) (1:2500) were diluted in 5% nonfat milk in TBST and incubated with the membrane overnight at 4°C. After washing, the membrane of primary antibody was incubated with HRP-conjugated goat anti-rabbit/mouse IgG secondary antibody (HY-P8001/HY-P8004) (1:5000) diluted in 5% nonfat milk in TBST for 1 h at room temperature. Protein bands were visualized using an Ultra High Sensitivity ECL detection kit (HY-K1005).

Background

  • Function

    CD38 is a glycoprotein found on the surface of many immune cells, including T cells, B cells and natural killer cells. Additionally, CD38 has been shown to be a bifunctional enzyme expressing both ADP-ribosyl cyclase and cADPR hydrolase activities. The role of CD38 in immune cells ranges from modulating cell differentiation to effector functions during inflammation, where CD38 may regulate cell recruitment, cytokine release, and NAD availability. In line with a role in inflammation, CD38 appears to also play a critical role in inflammatory processes during autoimmunity. Because of CD38 expression is highly upregulated on human plasma cells and especially on multiple myeloma cells.Targeting of CD38 is one of the most important and promising treatment avenues to help patients suffering from multiple myeloma.
