  • Function

    GRK3 is a GPCR kinase that phosphorylates agonist-occupied receptors and promotes β-arrestin-linked desensitization, internalization, and signaling control[1]. Mechanistically, GRK3 regulates receptor-specific pathways, including CXCR4 Ser-346/347 phosphorylation, where GRK3 knockdown produced the strongest reduction among GRK2, GRK3, and GRK6 knockdowns[2]. In disease models, GRK3 promoted prostate tumor growth and metastasis, regulated breast cancer migration and invasion, and drove gastric adenocarcinoma invasion through YAP1-associated transcriptional targets[3][4][5]. Compared with related isoforms, GRK3 differs from GRK2 in vivo because GRK2 loss causes embryonic lethality, whereas GRK3 deletion permits development but disrupts odorant receptor desensitization[6]. For experimental applications, LD2 specifically inhibited GRK3 but not the highly similar GRK2 in gastric adenocarcinoma models, while isochlorogenic acid A was identified as a GRK3 inhibitor in diabetic heart injury models[5][7].
