Verification Image
WB
  • Western blot analysis of extracts from Hela(lane 2(20μg), HEK293 (lane 3(20μg) and HepG2 (lane 4(20μg) using CDKN2A/p16INK4a (HY-P80992) Rabbit mAb. Proteins were transferred to a PVDF membrane and blocked with 5% non-fat milk in TBST for 2 hour at room temperature. The primary antibody (1/1000) and Loading control antibody (Beta Actin, HY-P80438, 1/10000) was used in 5% non-fat milk in TBST at 4°C overnight. Goat Anti-Mouse/Rabbit IgG-HRP Secondary Antibody (1/10000) was used for 1 hour at room temperature.

Background
Function:Acts as a negative regulator of the proliferation of normal cells by interacting strongly with CDK4 and CDK6. This inhibits their ability to interact with cyclins D and to phosphorylate the retinoblastoma protein
Subcellular Localization:Cytoplasm; Nucleus
Expression:
Tissue_specificity:It is widely expressed, but not detected in the brain or skeletal muscle. Isomer 3 is pancreas-specific.
Positive sample: HeLa cell lysate, HepG2 cell lysate, HEK-293 cell lysate, NIH:OVCAR-3 cell lysate, human ovary carcinoma tissue.
Isoforms & Post-Translational Modification:P42771 has 6 isomers: P42771-1: 16533 Da (predicted); P42771-2: 11215 Da (predicted); P42771-3: 12083 Da (predicted); Q8N726-1: 13903 Da (predicted); P42771-4: 17883 Da (predicted); Q8N726-2: 8731 Da (predicted).
Phosphorylation seems to increase interaction with CDK4
Subunit:Heterodimer with CDK4 or CDK6. Predominant p16 complexes contained CDK6. Interacts with CDK4 (both 'T-172'-phosphorylated and non-phosphorylated forms); the interaction inhibits cyclin D-CDK4 kinase activity. Interacts with ISCO2

Help & FAQs
  • Do most proteins show cross-species activity?

Species cross-reactivity must be investigated individually for each product. Many human cytokines will produce a nice response in mouse cell lines, and many mouse proteins will show activity on human cells. Other proteins may have a lower specific activity when used in the opposite species.
